The coffee at Koreatown’s Memorylook is as excellent as its matcha lattes, giant matcha brulee, and tiramisu doughnuts. There are actually two locations in LA, and they're fairly close to one another. One is located on Vermont Avenue, and the other is located at The Pearl on Wilshire Apartments. The specialty coffees are also a major hit, with the Liquid Gold matcha drink being a local favorite. The lavender syrup is herbal and sweet, and since it's served over ice, it's the perfect beverage for a warm summer day in LA. Habitat Coffee is located in Eagle Rock and specializes in far more than just coffee.

The modern coffee shop is situated right in the heart of Melrose Avenue in the Melrose Arts District, and you'll always find an array of locals and visitors popping in for a shot of espresso. Upon walking in, the first thing to greet guests is a wall of greenery that creates the outline of a sasquatch. The curved white booths are contemporary while still being inviting, and the shop can easily be used for working or hanging. Located on the Westside in the hip beach town of Venice, the original Menotti's Coffee Shop is a local gem that blends coffee with culture. However, the second Culver City location has become equally as popular, while the on-the-go sprinter van brings your favorite brew straight to you on wheels. Patrons are able to check where the van will be each week, but it can also be booked for parties and private events such as weddings or birthday celebrations.

Kodō Hotel is the brainchild of Boulevard Hospitality Group, which also owns the historic Japanese restaurant Yamashiro overlooking Hollywood, as well as such spots as Durango Cantina on Melrose and the three-level Cork & Batter in Inglewood. Boulevard recently launched Nice & Sweet, a new bakery and coffee shop in Hollywood, located at 7061 W. Whether you're in Los Feliz, Beverly Hills, or the Arts District in DTLA, Maru Coffee is not far. That said, all of the coffee is actually roasted at the DTLA location, and guests can always expect quality to remain a top priority.

Since then, after being featured on HBO's Insecure, the coffee shop is now backed by showrunner Issa Rae, who's helped Hilltop expand to Inglewood, Eagle Rock and Downtown. And Eagle Rock locations have plenty of tables and a few charging outlets apiece, while the Inglewood shop’s lofted couch seating is more conducive to casual morning or afternoon hangs. Endorffeine isn’t your cutesy, sun-dappled, Instagram-driven coffee shop—not that there isn’t a place for those, especially in L.A.

The white and yellow interior is modern yet aesthetic and warm, and to say the service is friendly would be an understatement. It is a cafe but also a community space that hosts special events. Staff serve incredible coffee beans sourced from around the globe and roast them at a site near the cafe.

There is also a modern appeal and style across each storefront, and there are always plenty of people working and gathering with their laptops throughout the entire week. The Highland Park and Studio City locations have more food menu items than the DTLA shop, but every kind of coffee lover and diner can find something to their liking. However, the secret menu at the Art's District location is a fun way to check out new and inventive coffees and beverages. The current secret menu item is hojicha green tea latte, which can be served either hot or cold. In regard to the food, Eightfold Coffee has everything from avocado toast to chia pudding with fresh fruit.

Don't be surprised if the baristas try to engage in conversation — that's sort of the brand. The butterfly latte is an Instagram favorite thanks to its vibrant coloring and use of blue butterfly pea flower. If you like sweeter coffees, there are plenty of great options on the menu. The zesty and slightly spicy "queen" avocado toast is also a great grab for breakfast.

That said, while trendy spots don't always serve the best food and drink, Alfred remains pretty consistent and hasn't lost sight of the importance of quality goods. The iced vanilla latte is one of the most popular drinks, but if you're looking for a bit of zen with your caffeine boost, order the iced lavender latte. Depending on which location you visit, Alfred also has breakfast items like bagels and cream cheese, breakfast sandwiches, yogurt and granola, and breakfast burritos.

Or check out its sister location, Woodcat Coffee, located farther down Sunset Boulevard in Echo Park. This grab-and-go coffee shop is a charming and quaint neighborhood spot that specializes in traditional espresso shots and concoctions. While most people order and take their goods on the go, there are a handful of tables inside where you can post up for an hour while you read a good book or catch up with a friend.

Directly across the street from Warner Music Group’s L.A. Offices, Kodō Hotel is tucked away on the second floor of a former Beaux Arts-style fire station that dates back to 1927. The husband and wife team have announced a second location in the same space as the Kaiser Baldwin Hills-Crenshaw Medical Offices (3782 W. Martin Luther King Jr. Boulevard), opening in late October. The Crenshaw/Baldwin Hills arm will have a massive outdoor seating area for those seeking medical service, but will undoubtedly attract locals from the adjacent residential neighborhood. Our bar has been a favorite spot for locals and visitors to enjoy a beer or cocktail and some fantastic local bands. Even though there wasn’t much space for dancing, you guys found a way!

The property’s ground-floor Kodō restaurant serves everything from sushi rolls and omakase platters to salads and main dishes like Uni-mentaiko Cream Pasta with cured egg yolk. It features minimalist meets earthy design in keeping with the aesthetic of the hotel, as well as an outdoor patio and a private dining room that’s located in the fire station’s former handball court. The New Orleans-style coffee is by far the most popular item on the menu. It's definitely on the sweeter side, but if you prefer to start your morning with a little sugar, it's the perfect drink no matter what time of year.

The Best Luxury Mother’s Day Gifts for Designer-Loving Moms

Making it a great spot for early risers and freelancers looking for a new all-day spot to work from. The coffee and espresso options are plentiful, ranging from a cafe au lait to an iced Americano. One of the coolest things about Habitat Coffee is the fact that it has more milk options than your standard coffee shop. In addition to the typical whole, nonfat, almond, and oak milks, Habitat Coffee also has rice milk, soy milk, and coconut milk. Though the coffee drinks are the specialty at Tilt, the matcha and other tea offerings are extensive. Located on the bottom floor of the Medallion apartments, Tilt is the place for coffee enthusiasts who crave good service and are in the mood for a strong single-origin cold brew.

Travel aficionados may recall that this spot had a brief life as another boutique hotel, the aptly named Firehouse Hotel, which enjoyed a brief run after opening in 2019, not far from such hotspot restaurants as Bestia and Bavel. Chimney Coffee House first opened in LA's Chinatown in 2010. If you're looking to see just how complex and dynamic coffee truly is, Chimney Coffee House is the place to go. The use of seasonal beans showcases differences in flavor and depth throughout the year. The small-batch roasting also ensures that quality remains intact as the beans are carefully tended to throughout the entire curating and roasting process.

The vine-covered walls set the scene on the outdoor patio for a productive yet romantic workday, and the quality of the coffee is equally as impressive. If you're looking for a sleek and minimalistic oasis to sip some quality brew, Maru Coffee should be at the top of your list. While the coffee should obviously be the main draw of any coffee shop, the design-forward interior and vibe at Maru are enough to make anyone want to visit. The atmosphere and ambiance are both calming and inspiring, and thankfully, there are three different locations in LA. Enjoy an elevated twist on classic coffee and pastries when you stop by Alchemist Coffee Project.

Prior to 1989, Station House Café occupied the space on the corner of Main and 3rd Street. That space recently became available, and we’ve relocated back to that original location — back to our roots. Prior to 1989, Station House Café occupied the space on the corner of Main and 3rd in downtown Point Reyes Station. That space became available, and we’re excited to share the news that we’ve relocated there — back to our roots.

The Oaxacan mocha is the most popular drink at BLVD MRKT’s Cafe Santo. This Oaxacan coffee bar is stationed right at the entry to the shipping container food hall and serves brioche french toast, breakfast sandwiches, and quiche as well. Kiff Kafe in West LA has secured our No. 1 spot as the best coffee shop in LA thanks to its transportive atmosphere, amazing service, and stellar selections. Located on West Pico Boulevard, visiting Kiff Kafe feels more like dining at a charming French bistro than it does an LA coffee shop.

This spacious and stylish coffee destination is busy no matter the time of day—no surprise, seeing as it’s one of Highland Park’s top shops to grab a cup and a croissant, now with a second location in Downtown L.A.'s Fashion District. The espresso blend rotates frequently, but teas and specialty coffee drinks remain more or less the same. We love the Figueroa, made with sweetened condensed milk, espresso, milk and cinnamon, topped with a Maria Cookie. No matter which location you visit, the beans at Boy & the Bear get roasted three times a week, which makes a major difference in the quality of each cup. All four cafés also have Wi-Fi, charging outlets and at least a few couches, and the Redondo Beach flagship has the most space and outdoor seating. A cup of joe is perhaps the most classic and universally loved morning ritual shared by people all across the globe.

While some people refuse to give up on their Starbucks or Dunkin' Donuts order, branching out and trying locally owned and independent coffee shops can open your eyes to the magical world of quality coffee. Whether you stick with a simple cold brew to get your day going or you prefer a milky latte, coffee is far more complex than most people realize. Hyper curated and hyper technical, Highland Park's Kumquat Coffee is a caffeine lover's paradise. They also offer fresh pastries and breakfast burritos, just in case all that caffeine isn't fuel enough. In 2018, Yonnie Hagos and Ajay Relan first opened in the historically Black neighborhood of View Park-Windsor Hills, hoping to help locals connect, caffeinate and continue the climb towards success.

Coffee drinks are available any time of day and are best enjoyed on the pleasant patio if the weather permits. If headed to Community Goods during busy hours, settle into the line with a book or a podcast. The queue is consistent, as are the stellar coffee drinks like the einspänner latte with sweetened cream, cocoa powder, double espresso, and a pinch of salt. Between the funky architecture and bright and airy interior, Coffee for Sasquatch is a design enthusiast's dream.

Add in the blue butterfly pea flower matcha latte, horchata cold brew, and a range of specialty beans, and this is one cafe you don't want to miss. Bloom & Plume is open seven days a week, ensuring that loyal regulars can get their favorite cup of brew each and every morning. This Black- and family-owned shop has been a beloved neighborhood business since its opening in 2019. Though you can easily get traditional lattes and caffeinated beverages, Bloom & Plume is most popular for its more inventive beverages that bring a fresh approach to the world of coffee. The Channel Orange latte has hints of citrus, making it taste similar to a creamsicle.

CRISPR-GPT: An LLM Agent for Automated Design of Gene-Editing Experiments

design based research

The first difference between an experimental approach and design-based research regards the role participants play in the experiment. In an experimental approach, the researcher is responsible for making all the decisions about how the experiment will be implemented and analyzed, while the instructor facilitates the experimental treatments. In design-based research, both researchers and instructors are engaged in all stages of the research from conception to reflection (Collins et al., 2004). In BER, a third condition frequently arises wherein the researcher is also the instructor.

Design-Based Research Invites Using Mixed Methods to Analyze Data

However, over generations of research, this connection has been all but forgotten, and DBR, although similarly inspired by the early efforts of Simon, Archer, and Jones, has developed into an isolated and discipline-specific body of design research, independent from its interdisciplinary cousin. Given the conceptual framework, theoretical and research questions, and sociopolitical interests at play, researchers may undertake this, and subsequent steps in the process, on their own, or in close collaboration with the communities and individuals in the situated contexts in which the design will unfold. As such, across iterations of DBR, and with respect to the ways DBR researchers choose to engage with communities, the origin of the design will vary, and might begin in some cases with theoretical questions, or arise in others as a problem of practice (Coburn & Penuel, 2016), though as has been noted, in either case, theory and practice are necessarily linked in the research. By employing this methodology from the learning sciences, biology education researchers can enrich our current understanding of what is required to help biology students achieve their personal and professional aims during their college experience.

Methodological Issues in the Design-Based Research Paradigm

Research subjects are co-workers with the researcher in iteratively pushing the study forward. In DBR, researchers assume the roles of “curriculum designers, and implicitly, curriculum theorists” (Barab & Squire, 2004, p.2). As curriculum designers, DBR researchers come into their contexts as informed experts with the purpose of creating, “test[ing] and refin[ing] educational designs based on principles derived from prior research” (Collins et al., 2004, p. 15). These educational designs may include curricula, practices, software, or tangible objects beneficial to the learning process (Barab & Squire, 2004). As curriculum theorists, DBR researchers also come into their research contexts with the purpose to refine extant theories about learning (Brown, 1992). Ultimately, however, these responses are likely to always be insufficient as evidence of rigor to some, for the root dilemma is around what “counts” as education science.

Experimental research designs

Practitioners are able to benefit from research when they see how the research can inform and improve their designs and practices. Some practitioners believe that educational research is often too abstract or sterilized to be useful in real contexts (The Design-Based Research Collective, 2002). DBR originated as researchers like Allan Collins (1990) and Ann Brown (1992) recognized that educational research often failed to improve classroom practices. They perceived that much of educational research was conducted in controlled, laboratory-like settings.

Case study

design based research

However, the iterative and multi-tasking nature of a DBR process may not be well-suited to empirically testing or building theory. According to Hoadley (2004), “the treatment’s fidelity to theory [is] initially, and sometimes continually, suspect” (p. 204). This suggests that researchers, despite intentions to test or build theory, may not design or implement their solution in alignment with theory or provide enough control to reliably test the theory in question.

Specifically, in most DBR, this means a focus on “intermediate-level” theories of learning, rather than “grand” ones. In essence, DBR does not contend directly with “grand” learning theories (such as developmental or sociocultural theory writ large) (diSessa, 1991). Rather, DBR seeks to offer constructive insights by directly engaging with particular learning processes that flow from these theories on a “grounded,” “intermediate” level. This is not, however, to say DBR is limited in what knowledge it can produce; rather, tinkering in this “intermediate” realm can produce knowledge that informs the “grand” theory (Gravemeijer, 1994). For example, while cognitive and motivational psychology provide “grand” theoretical frames, interest-driven learning (IDL) is an “intermediate” theory that flows from these and can be explored in DBR to both inform the development of IDL designs in practice and inform cognitive and motivational psychology more broadly (Joseph, 2004). Put in other terms, even as DBR researchers may choose heterogeneous methods for data collection, data analysis, and reporting results complementary to the ideological and sociopolitical commitments of the particular researcher and the types of research questions that are under examination (Bell, 2004), a shared epistemic commitment gives the methodology shape.

This call for more scholarly discussion and practice resonated with designers across disciplines in design and engineering (Buchanan, 2007; Cross, 1999; Cross, 2007; Friedman, 2003; Jonas, 2007; Willemien, 2009). IDR sprang directly from this early movement and has continued to gain momentum, producing an interdisciplinary body of research encompassing research efforts in engineering, design, and technology. Design-Based Research (DBR) is one of the most exciting evolutions in research methodology of our time, as it allows for the potential knowledge gained through the intimate connections designers have with their work to be combined with the knowledge derived from research. These two sources of knowledge can inform each other, leading to improved design interventions as well as improved local and generalizable theory.

First of all, it is necessary to think of the best way to operationalize the variables that will be measured, as well as which statistical methods would be most appropriate to answer the research question. Thus, the researcher should consider what the expectations of the study are as well as how to analyze any potential results. Finally, in an experimental design, the researcher must think of the practical limitations including the availability of participants as well as how representative the participants are to the target population.

Understanding these shared features, in the context and terms of the methodology itself, help us to appreciate what is involved in developing robust and thorough DBR research, and how DBR seeks to make strong, meaningful claims around the types of research questions it takes up. As with any methodological approach, there can be challenges to implementing design-based research. In this new study, researchers leverage modern predictors' utilization of a Path History Register (PHR) to index prediction tables. The PHR records the addresses and precise order of the last 194 taken branches in recent Intel architectures.

This suggested that students had developed nascent flux conceptual frameworks after experiencing the instructional tools but could use more support to realize the broad applicability of this principle. Also, although our cross-sectional interview approach demonstrated how students’ ideas, overall, could change after experiencing the instructional tools, it did not provide information about how a student developed flux reasoning. A major goal of design-based research is producing meaningful interventions and practices. Within educational research these interventions may “involve the development of technological tools [and] curricula” (Barab & Squire, 2004, p. 1). But more than just producing meaningful educational products for a specific context, DBR aims to produce meaningful, effective educational products that can be transferred and adapted (Barab & Squire, 2004).

Here, using large language models (LLMs) trained on biological diversity at scale, we demonstrate the first successful precision editing of the human genome with a programmable gene editor designed with AI. To achieve this goal, we curated a dataset of over one million CRISPR operons through systematic mining of 26 terabases of assembled genomes and meta-genomes. We demonstrate the capacity of our models by generating 4.8x the number of protein clusters across CRISPR-Cas families found in nature and tailoring single-guide RNA sequences for Cas9-like effector proteins. Several of the generated gene editors show comparable or improved activity and specificity relative to SpCas9, the prototypical gene editing effector, while being 400 mutations away in sequence. Finally, we demonstrate an AI-generated gene editor, denoted as OpenCRISPR-1, exhibits compatibility with base editing. We release OpenCRISPR-1 publicly to facilitate broad, ethical usage across research and commercial applications.

Qualitative design-based research (DBR) first emerged in the learning sciences field among a group of scholars in the early 1990s, with the first articulation of DBR as a distinct methodological construct appearing in the work of Ann Brown (1992) and Allan Collins (1992). For learning scientists in the 1970s and 1980s, the traditional methodologies of laboratory experiments, ethnographies, and large-scale educational interventions were the only methods available. The laboratory, or laboratory-like settings, where research on learning was at the time happening, was divorced from the complexity of real life, and necessarily limiting. Alternatively, most ethnographic research, while more attuned to capturing these complexities and dynamics, regularly assumed a passive stance1 and avoided interceding in the learning process, or allowing researchers to see what possibility for innovation existed from enacting nascent learning theories.

Proponents of DBR believe that conducting research in context, rather than in a controlled laboratory setting, and iteratively designing interventions yields authentic and useful knowledge. Sasha Barab (2004) says that the goal of DBR is to “directly impact practice while advancing theory that will be of use to others” (p. 8). This implies “a pragmatic philosophical underpinning, one in which the value of a theory lies in its ability to produce changes in the world” (p. 6).

In this case, if the research questions being investigated produce generalizable results that have the potential to impact teaching broadly, then this is consistent with a design-based research approach (Cobb et al., 2003). However, when the research questions are self-reflective about how a researcher/instructor can improve his or her own classroom practices, this aligns more closely with “action research,” which is another methodology used in education research (see Stringer, 2013). There is, of course, much more nuance to these models, and each of these models (formative interventions, Change Laboratories, social design experiments, and participatory design research) might itself merit independent exploration and review well beyond the scope here. Indeed, there is some question as to whether all adherents of these CHAT design-based methodologies, with their unique genealogies and histories, would even consider themselves under the umbrella of DBR.
